EPCO-45. EXTRACHROMOSOMAL DNA PROMOTES ONCOGENE AMPLIFICATION ACROSS MULTIPLE PEDIATRIC CANCER TYPES

چکیده
Abstract Extrachromosomal circular DNA (ecDNA) is an important driver of aggressive cancers. Its frequency and impact on pediatric cancers has yet to be fully described. To determine the ecDNA prevalence in various cancer types, we accessed whole genome sequencing (WGS) data Children’s Brain Tumor Network (CBTN) St. Jude cloud genomics platforms utilized bioinformatics tools Amplicon Architect (AA) Classifier (AC) reconstruct sequence structures. We then identified which genes were amplified how correlated with survival, using Kaplan Meier curves. Different types noted have varying highest detection high-grade gliomas (30% cases). In analyzing matched diagnostic/relapse relapse/progression pairs, was evolve over time progression disease. Our preliminary results demonstrate that prevalent cancer, can contain known potential novel oncogenes associated poorer outcomes across many different types.

Oncogene amplification in pediatric brain tumors.
Despite a considerable amount of information concerning chromosomal and molecular abnormalities found in gliomas in adults, relatively little is known regarding these abnormalities in pediatric brain tumors. We have analyzed DNA from 37 primary brain tumors and 4 tumor-derived cell lines for oncogene amplification. Probes utilized represent 11 known oncogenes (erbB1, gli, neu, myc, L-myc, N-myc...
متن کاملAmplification of a Zygosaccharomyces bailii DNA Segment in Wine Yeast Genomes by Extrachromosomal Circular DNA Formation
We recently described the presence of large chromosomal segments resulting from independent horizontal gene transfer (HGT) events in the genome of Saccharomyces cerevisiae strains, mostly of wine origin. We report here evidence for the amplification of one of these segments, a 17 kb DNA segment from Zygosaccharomyces bailii, in the genome of S. cerevisiae strains. The copy number, organization ...
متن کاملSMURF1 Amplification Promotes Invasiveness in Pancreatic Cancer
Pancreatic cancer is a deadly disease, and new therapeutic targets are urgently needed. We previously identified DNA amplification at 7q21-q22 in pancreatic cancer cell lines. Now, by high-resolution genomic profiling of human pancreatic cancer cell lines and human tumors (engrafted in immunodeficient mice to enrich the cancer epithelial fraction), we define a 325 Kb minimal amplicon spanning S...
متن کاملHer-2/neu oncogene amplification in clinically localised prostate cancer.
AIM To examine the incidence of Her-2/neu oncogene amplification in clinically localised prostate cancer using in situ hybridisation. METHODS One hundred and seventeen patients, who had undergone radical prostatectomy, were identified and in situ hybridisation was performed on formalin fixed, paraffin wax embedded tissue using the Quantum Appligene probe for Her-2/neu. The enzyme peroxidase w...
